Human Fibroblast growth factor 21 - cat. no. CYT-24 | |
Description: | Human FGF-21 is a secreted growth factor that is a member of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. Recombinant human FGF-21 is a 19.5 kDa protein containing 182 amino acid residues. Human FGF-21 is purified by proprietary ion exchange chromatographic techniques. |

Lot Number: | |
Quantity Shipped: | |
Physical Appearance: | White lyophilized (freeze-dried) powder. |
Formulation: | The protein was lyophilized from a concentrated (0.8 mg/ml) solution with 0.4 mg/ml of NaHCO3 previously adjusted pH 8. |
Solubility: | It is recommended to reconstitute the lyophilized human FGF-21 in 0.4% NaHCO3 or water, not less than 100µg/ml, which can then be further diluted to other aqueous solutions, preferably in a presence of a carrier protein such as BSA or similar. |
Stability: | Lyophilized hFGF-21 although stable at room temperature for 3 weeks, should be stored desiccated below -18C. Upon reconstitution and filter sterilization hFGF-21 can be stored at 4C, pH 8 for up to 4 weeks. For long term storage and more diluted solutions (but not for in vivo experiments) it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles. |
Purity: | Greater than 98.0% as determined by: (a) Gel filtration at non denaturing conditions using 25 nM Tris-HCl + 300 mM NaCl, pH 8. (b) Analysis by reducing and non-reducing SDS-PAGE gel. |
Amino Acid Sequence: | The sequence of the first eight N-terminal amino acids was determined and was found to be Ala-His-Pro-Ile-Pro-Asp-Ser-Ser. |
Dimers and Aggregates: | Less than 2% as determined by gel filtration chromatography. |
Biological Activity: | hFGF21 lowered body weight in diet-induced obese mice when delivered via mini-osmotic pumps. |
Endotoxin: | Less than 0.05 ng/µg (0.5 EU/µg) of hFGF-21. |
Protein content: | Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.74 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the DNAman computer analysis program of protein sequences |
