Description: Recombinant Gilthead Seabream (Sparus aurata) growth hormone (gsGH) produced in E. coli (21.4 K) is a single, non-glycosylated, polypeptide chain containing 188 amino acids and having a molecular mass of ~ 21.392 kDa. Recombinant Gilthead Seabream growth hormone was purified by chromatographic techniques, according to Ben-Atia et al., General and Comparative Endocrinology 113, 155–164 (1999).

Physical Appearance: White lyophilized (freeze-dried) powder. 
Formulation: The recombinant Gilthead Seabream growth hormone was lyophilized from a concentrated (1mg/ml) solution with 0.02% NaHCO3. 
Solubility: It is recommended to reconstitute the lyophilized recombinant Gilthead Seabream growth hormone in sterile 0.4% NaHCO3 adjusted, not less than 100µg/ml, which can then be further diluted to other aqueous solutions. 
Stability: Lyophilized recombinant Gilthead Seabream growth hormone although stable at room temperature for several weeks, should be stored desiccated below -18 C. Upon reconstitution of recombinant Gilthead Seabream growth hormone at > 0.1 mg/ml and up to 2 2 mg/ml and filter sterilization recombinant Gilthead Seabream growth hormone can be stored at +4C. 
Purity: The purity of recombinant Gilthead Seabream growth hormone is greater than 98.0% as determined by:
. (a) Analysis by reducing and non-reducing SDS-PAGE gel.
(b) Gel-filtration chromatography under non denaturing conditions. 
Amino Acid Sequence: The sequence of the first ten N-terminal amino acids of recombinant Gilthead Seabream growth hormone was determined and was found to be Ala-Gln-Pro-Ile-Thr-Asp-Gly-Gln-Arg-Leu.  
Dimers and Aggregates: The purified recombinant Gilthead Seabream growth hormone consists of > 97% monomers as determined by gel-filtration chromatography. 
Biological Activity: Binding assays of the 125I-labeled PLR’s recombinant Gilthead Seabream growth hormone to dolphin fish liver microsomal fraction resulted in high specific binding characterized by a Ka of 1.93 nM and a Bmax of 540 fmol/mg microsomal fraction protein. Recombinant Gilthead Seabream growth hormone like ovine placental lactogen, exhibited growth-stimulating activity when applied orally to S.aurata larvae or intraperitoneally to juvenile fish. 
Endotoxin: Less than 0.1 ng/µg (IEU/µg) of recombinant Gilthead Seabream growth hormone.  
Protein content: Protein quantitation of recombinant Gilthead Seabream growth hormone was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.72 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the DNAman computer analysis program of protein sequences. 
Usage: The recombinant Gilthead Seabream growth hormone is offered by PLR for laboratory research
