Description:
IL10 is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in inhibition of immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract.

Formulation:
The protein was lyophilized from a concentrated (1mg/ml) solution containing 0.02-0.03% of NaHCO3 and glycine.
Solubility:
Lyophilized Interleukin 10 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL10 should be stored sterile at 4°C between 7-14 days and for future use below -18°C.

Stability:
It is recommended to reconstitute the lyophilized hIL 10 in sterile water or 0.4% NaHCO3 adjusted tp pH 8-9, not less than 100µg/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein such as BSA or HSA.

Amino Acid Sequence:
The sequence of the first four N-terminal amino acids was determined and was found to be Ala-Ser-Pro-Gly-Gln.
Dimers and Aggregates:
The purified hIL 10 consists of > 90% dimers as determined by gel-filtration chromatography under non reducing conditions.
Biological Activity:
PLR’s hIL 10 is fully biologically active as evidenced by its ability to induce proliferation of IM/9 cells in presence of IL 3.

Less than 0.0025 ng/µg (0.025 IEU/µg) of hIL 10
Protein content:
Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.425 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
