Mouse leptin D23L mutant - Cat no. LEP-41 GENE ID 16846 NEW

Description:
Recombinant mouse leptin mutant D23L, one polypeptide chain containing 146 amino acids and additional Ala at N-terminus with molecular mass od ! 16 kDa was purified by propertiary chromatographic techniques similarly to that described in Salomon et al. (2006) in Protein Expression and Purification 47, 128-136
Source:
E. coli
Physical Appearance:
White lyophylized poeder
Formulation:
The recombinan protein was lyophilized from a concentrated (1mg/ml) solution with 0.0045mM NaHCO3.It is recommended to reconstitute the lyophilized recombinant mouse leptin in sterile water or 0.4% NaHCO3 adjusted tp pH 8-9, not less than 100µg/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein.
Solubility:
It is recommended to reconstitute the lyophilized recombinant mouse leptin in sterile water or 0.4% NaHCO3 adjusted tp pH 8-9, not less than 100µg/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein.
Stability:
Lyophylized recombinant mouse leptin mutant though is stable at room temperature for several weeks, should be stored velow -18 C. Upon reconstitution at > 0.1 mg/ml and up to 2.0 mg/ml and filter sterilization can be stored at 4 C for 2-3 months.
Purity:
The purity of recombinant mouse leptin D23L mutant is > 95% as determined by:
(1) Gel filtration (SEC) analysis
(2) Analysis by reducing and non-reducing SDS-PAGE gel.
Amino Acid Sequence:
The sequence of the first five N-terminal amino acids is Ala-Val-Pro-Ile-Gln
Dimers and Aggregates:
The purified mouse leptin D23L mutan consist of >95% monomers as determined by SEC.
Biological Activity:
The biological activity of mouse leptin mutant was analyzed by inducing proliferation of BA/F3 cells stably transfercted with the long for of human leprin receptor but its affinity toward this receptor was ~ 25-fold higher compared to non-mutated mouse leptin.
Endotoxin:
Less 0.05 ng/microgram of mouse leptin mutatnt
Protein content:
Protein quantification of recombinant mouse leptin D23L mutant was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.2 for 0.1% solution. This value was calculated by DNA man program.
Usage:
Recombinant mouse leptin mutant D23L is offered by PLR for laboratory research
